Pill Crush Testing for Pharmaceutical Applications

The Challenge

Pill crush testing is performed to evaluate the compressive properties of tablets and ensure they can withstand the range of forces encountered during manufacturing, packaging, shipping, and handling. A typical test requires a pill to be compressed to failure or compressed to a defined force level and checked for damage or puncture.

While the test itself is relatively straightforward, fixture selection is critical. Using a compression platen or probe that is incorrectly sized can cause the pill to slip out from under load or result in uneven force distribution across the pill's surface — both of which compromise the accuracy and repeatability of the result. Test speed is equally important, as pills may be subjected to very different rates of force application depending on the stage of the manufacturing and distribution process being simulated.

Pill crush testing is commonly performed in accordance with pharmacopoeial standards including USP and Ph. Eur., which define requirements for tablet hardness and friability.

Test Speed and Simulation Conditions

To accurately simulate the range of conditions a pill may encounter — from high-speed packaging machinery to manual handling — we recommend performing pill crush tests across a range of speeds. Using choice inputs and linked values in Bluehill® Universal software, operators can select a simulation condition such as packaging or handling from a simple drop-down menu. The software automatically adjusts the test speed behind the scenes, eliminating the need for manual reconfiguration and reducing the risk of operator error between test conditions.

Fixture Selection

Correct fixture selection is essential for accurate and repeatable pill crush testing. We recommend using at least a 2-inch compression platen on the base of the system to provide a stable, level surface that fully supports the pill and prevents it from sliding under load. On the upper fixture, a smaller probe sized to closely match the pill's diameter ensures that the force is applied consistently to the correct area of the tablet surface. Using a probe that is too large risks distributing the load unevenly, while a probe that is too small may concentrate force in a way that does not reflect real-world conditions.

Recommended Load Cell

For most pill crush applications, a low-capacity load cell in the range of 50 N to 500 N is appropriate, depending on the hardness of the tablet being tested. Instron's 2580 Series load cells offer accuracy down to 1/1000th of the load cell capacity, ensuring reliable measurement even at the lower end of the force range.
